RNAi was used to analyze the effects of suppressing <i>rpn-13 </i>and codepleting <i>rpn-13</i> and <i>wee-1.3, </i>on C.elegans fertility<i>.</i> Strain IT1877 contains a mutated ubiquitin fused to a GFP, and therefore monitors proteolytic activity of the 26S Proteasome. If gene depletions cause 26S proteasome dysfunction, the degree of dysfunction can be quantified by measuring the intensity of the emitted signal. A brighter GFP signal indicates proteasome dysfunction, while low to minimal GFP signal means the proteasome is functioning normally.
